چکیده

Title of Document: BASELINE ADJUSTMENT FOR ORDINAL COVARIATES BY INDUCING A PARTIAL ORDERING IN RANDOMIZED CLINICAL TRIALS. Na An, Master of Arts, 2007 Directed By: Professor Paul J. Smith Department of Mathematics In two-armed randomized clinical trials (RCTs) designed to compare a new treatment with a control, a key endpoint is often measured and analyzed both at baseline and after treatment for two groups. More powerful and precise statistical inferences are possible once the between-group comparisons have been adjusted for covariates. In this thesis we propose a new method for ordered categorical outcomes which adjusts for baseline without relying on any specific assumptions on the data generating process. Based on baseline and post-treatment values, data are composed of counts of patients who have improved from one category to another, stayed the same or deteriorated. Not all patterns are comparable. Hence, the ordering is only partial. We develop an approach to test the treatment effects based on comparing each observation in one group to each observation in the other group to which it is comparable. The power comparisons of this test with four common approaches are conducted in our simulation study. BASELINE ADJUSTMENT FOR ORDINAL COVARIATES BY INDUCING A PARTIAL ORDERING IN RANDOMIZED CLINICAL TRIALS.

Innovative Strategies for Strengthening Interpretability

Analysis of covariance serves two main purposes in the analysis of clinical trials. It provides for a more powerful statistical test through variance reduction for comparison of treatment groups; and it addresses comparisons between randomized groups with adjustment for random imbalance of covariates. Certain methods of covariance analysis such as logistic regression for ordinal response data a...

Covariate adjustment for two-sample treatment comparisons in randomized clinical trials: a principled yet flexible approach.

There is considerable debate regarding whether and how covariate-adjusted analyses should be used in the comparison of treatments in randomized clinical trials. Substantial baseline covariate information is routinely collected in such trials, and one goal of adjustment is to exploit covariates associated with outcome to increase precision of estimation of the treatment effect. However, concerns...

Bayesian Clustering with Regression

We propose a model for covariate-dependent clustering, i.e., we develop a probability model for random partitions that is indexed by covariates. The motivating application is inference for a clinical trial. As part of the desired inference we wish to define clusters of patients. Defining a prior probability model for cluster memberships should include a regression on patient baseline covariates...

Modeling Paired Ordinal Response Data

About 25 years ago, McCullagh proposed a method for modeling univariate ordinal responses. After publishing this paper, other statisticians gradually extended his method, such that we are now able to use more complicated but efficient methods to analyze correlated multivariate ordinal data, and model the relationship between these responses and host of covariates. In this paper, we aim to...
